Different responses of the GlnB and GlnZ proteins upon in vitro uridylylation by the Azospirillum brasilense GlnD protein BJMBR
Araújo,L.M.; Huergo,L.F.; Invitti,A.L.; Gimenes,C.I.; Bonatto,A.C.; Monteiro,R.A.; Souza,E.M.; Pedrosa,F.O.; Chubatsu,L.S..
Azospirillum brasilense is a diazotroph found in association with important agricultural crops. In this organism, the regulation of nitrogen fixation by ammonium ions involves several proteins including the uridylyltransferase/uridylyl-removing enzyme, GlnD, which reversibly uridylylates the two PII proteins, GlnB and GlnZ, in response to the concentration of ammonium ions. In the present study, the uridylylation/deuridylylation cycle of A. brasilense GlnB and GlnZ proteins by GlnD was reconstituted in vitro using the purified proteins. The uridylylation assay was analyzed using non-denaturing polyacrylamide gel electrophoresis and fluorescent protein detection. Our results show that the purified A. brasilense GlnB and GlnZ proteins were uridylylated by...

Residual effects of successive exposure of soybean Bradyrhizobium strains to aluminium on solid defined medium PAB
Campo,Rubens José; Wood,Martin.
The aim of these studies was to investigate whether residual toxic effects of exposing soybean root nodule bacteria to Al in a solid defined media (SDM) alter tolerance to Al, survival, sensitivity to antibiotics, N2 fixation effectiveness and genetic diversity of Bradyrhizobium strains. After being exposed four times to Al, strains showed variation in Al tolerance but there was no evidence of change in their original Al tolerance, sensitivity to the antibiotics or genetic diversity. Exposure of Bradyrhizobium strains to SDM plus Al did not alter biological N2 fixation effectiveness of five strains. Strain SEMIA 587 showed a reduction in its N2 fixation effectiveness but it seems that it was just a superficial toxic effect because one single passage...

Subtractive library of soybean roots in response to inoculation with Bradyrhizobium japonicum. Infoteca-e
CARVALHO, G. A. B. de; BATISTA, J. S. S.; MARCELINO, F. C.; HUNGRIA, M..
ABSTRACT: Soybean [Glycine max (L.) Merr.] is the most important legume cropped worldwide, with an economic value attributed mainly to the high protein content of the grain, which, in turn, demands a heavy supply of nitrogen. An environmentally friendly source of nitrogen at low cost to farmers is represented by the biological fixation of atmospheric nitrogen that takes place is association with symbiotic bacteria. For the soybean, the association occurs mainly with bacteria belonging to the species Bradyrhizobium japonicum and Bradyrhizobium elkanii, but our knowledge about gene expression associated with the symbiosis is still poor. This work - part of an effort by the Soybean Genome Consortium (Genosoja)?was conducted with the aim of achieving better...

Tricalcium phosphate solubilization and nitrogen fixation by newly isolated Aneurinibacillus aneurinilyticus CKMV1 from rhizosphere of Valeriana jatamansi and its growth promotional effect BJM
Chauhan,Anjali; Guleria,Shiwani; Balgir,Praveen P.; Walia,Abhishek; Mahajan,Rishi; Mehta,Preeti; Shirkot,Chand Karan.
Abstract Aneurinibacillus aneurinilyticus strain CKMV1 was isolated from rhizosphere of Valeriana jatamansi and possessed multiple plant growth promoting traits like production of phosphate solubilization (260 mg/L), nitrogen fixation (202.91 nmol ethylene mL-1 h-1), indole-3-acetic acid (IAA) (8.1 µg/mL), siderophores (61.60%), HCN (hydrogen cyanide) production and antifungal activity. We investigated the ability of isolate CKMV1 to solubilize insoluble P via mechanism of organic acid production. High-performance liquid chromatography (HPLC) study showed that isolate CKMV1 produced mainly gluconic (1.34%) and oxalic acids. However, genetic evidences for nitrogen fixation and phosphate solubilization by organic acid production have been reported first time...

Azospirillum sp . inoculation in wheat, barley and oats seeds greenhouse experiments BABT
Dalla Santa,Osmar R.; Hernández,Ramona Fernández; Alvarez,Gergina L. Michelena; Ronzelli Junior,Pedro; Soccol,Carlos Ricardo.
The aim of the present work was to evaluate the inoculation effects of Azospirillum sp. RAM-7 strain, in seeds of wheat, barley and oats cultures on yield and total nitrogen content of the grains in greenhouse experiments. Seeds inoculation with Azospirillum sp. RAM-7 strain enhanced grain yields, even so, the responses vary among the evaluated cultures. For wheat, significant increases were obtained when the inoculation was associated to 100% of the recommended nitrogen, although the treatment that received only inoculation produced 7.4% above the control, however this difference wasn't significant. For barley, the presence of the inoculant substituted 20% the recommended nitrogen fertilization. For oats the inoculation with Azospirillum sp. RAM-7 didn't...

Cyanobacterial nitrogenases: phylogenetic diversity, regulation and functional predictions Genet. Mol. Biol.
Esteves-Ferreira,Alberto A.; Cavalcanti,João Henrique Frota; Vaz,Marcelo Gomes Marçal Vieira; Alvarenga,Luna V.; Nunes-Nesi,Adriano; Araújo,Wagner L..
Abstract Cyanobacteria is a remarkable group of prokaryotic photosynthetic microorganisms, with several genera capable of fixing atmospheric nitrogen (N2) and presenting a wide range of morphologies. Although the nitrogenase complex is not present in all cyanobacterial taxa, it is spread across several cyanobacterial strains. The nitrogenase complex has also a high theoretical potential for biofuel production, since H2 is a by-product produced during N2 fixation. In this review we discuss the significance of a relatively wide variety of cell morphologies and metabolic strategies that allow spatial and temporal separation of N2 fixation from photosynthesis in cyanobacteria. Phylogenetic reconstructions based on 16S rRNA and nifD gene sequences shed light on...

Soil nutrient amendment with leaves of varying quality. Infoteca-e
FERNANDES, T. P.; SILVA, R. B.; SILVA JUNIOR, J. P. da; LEHMANN, J..
Study on the impact of nitrogen and phosphorus fertilization combined with litter amendment on the microbial biomass ofa xanthic Ferralso. (0cm-5cm and 10cm-20cm). First peliminary results after 18 days of incubation showed that soil amended with P tended to have higher microbial C than soils fertilized with N or the control. This result indicates that nitrogen is probably not limiting microbial biomass growth the most , but rather P.

Nitrogen fixation in the eastern Atlantic reaches similar levels in the Southern and Northern Hemisphere ArchiMer
Fonseca-batista, Debany; Dehairs, Frank; Riou, Virginie; Fripiat, Francois; Elskens, Marc; Deman, Florian; Brion, Natacha; Queroue, Fabien; Bode, Maya; Auel, Holger.
Euphotic layer dinitrogen (N-2) fixation and primary production (PP) were measured in the eastern Atlantic Ocean (38 degrees N-21 degrees S) using N-15(2) and C-13 bicarbonate tracer incubations. This region is influenced by Saharan dust deposition and waters with low nitrogen to phosphorus (N/P) ratios originating from the Subantarctic and the Benguela upwelling system. Depth-integrated rates of N-2 fixation in the north (0 degrees N-38 degrees N) ranged from 59 to 370 mu mol N m(-2) d(-1), with the maximal value at 19 degrees N under the influence of the northwest African upwelling. Diazotrophic activity in the south (0 degrees S-21 degrees S), though slightly lower, was surprisingly close to observations in the north, with values ranging from 47 to 119...
